چکیده

گونه‌­های جنس اسیدیتیو باسیلوس (Acidithiobacillus) در فرایند فروشویی زیستی فلزهای مختلف از جمله اورانیم، مس، نیکل استفاده می‌­شود. هدف از این بررسی، تدوین روش مناسب برای نگه‌­داری باکتری­‌های بومی جداسازی شده دخیل در فروشویی زیستی اورانیم با امکانات قابل دسترس است. در این پژوهش، باکتری­‌های دخیل در فروشویی زیستی اورانیم موجود در آزمایشگاه بیولیچینگ سازمان انرژی اتمی ایران پس از کشت و اطمینان از خالص بودن، در دماهای 4 و 800C - نگه‌­داری شدند. آن­گاه، بعد از گذشت 6 ماه از زمان نگه‌داری، باکتری‌­ها از نظر تغییر pH، Eh، رشد و ویژگی‌­های بیوشیمیایی بررسی، و با زمان قبل از نگه­‌داری مقایسه شدند. نتایج به دست آمده از این پژوهش حاکی از آن است که نگه‌­داری باکتری­‌ها در این مدت و در این دماها، هیچ­گونه اثری بر روی ویژگی­‌های بیوشیمیایی آن­ها نگذاشته است. به علاوه، منحنی­‌های به دست آمده از روندهای تغییر pH، Eh و رشد باکتری­‌های مورد نظر، تغییر معنی­‌داری در زمان قبل و بعد از نگه‌­داری نشان ندادند. این نتیجه بیان­‌کننده‌­ی آن  است که نگه‌­داری باکتری‌­های بومی در دماهای 4 و 800C-، عملکرد مؤثر آنها را حفظ می­‌کند و باکتری­‌ها پس از 6 ماه توانایی استفاده­‌ی مؤثر در فرایند فروشویی زیستی را هم­‌چنان دارند.

Following up of the Function of Acidithiodabacillus Ferrooxidans and Acidithiobacillus Thiooxidans Involved in Uranium Bioleaching After Preservation at Low Temperatures

چکیده [English]

Acidithiobacillus species is used in bioleaching process of metals such as uranium, copper, nickel. The purpose of this study was to develop an effective protocol using facilities available for preservation of isolated native bacteria involved in uranium bioleaching. In this study, the bacteria involved in uranium bioleaching AEOI  Laboratory were cultured, purified and stored at 4 and -80°C. Then, variation of pH, Eh, growth and biochemical characteristics were examined after 6 months of storage and compared with the bacteria before using the preservation methods. The results indicated that the preservation of the bacteria within 6 months at selective temperatures did not have any effect on the biochemical characteristics of the bacteria. In addition, no significant changes were observed in the variation of pH, Eh and growth of bacteria, before and after the preservations. The results suggested that 6 months preservations of native bacteria at 4 and -80°C, had no effect on the ability of bacteria in bioleaching process.
